Determination of Pectinase Enzyme using Streptomyces, Isolated from Fadama Soil Sample, Birnin Kebbi, Kebbi State, Nigeria

Abstract

Pectinase are a versatile class of enzymes owning to their ability to perform a specific range of biotransformations. The aim of this research work is to determine the activity of lipase enzyme from soil streptomyces and optimize the culture condition for maximum activity. The aim of this research is to produce lipase enzyme from soil Streptomyces and optimize the production media for maximum production of the enzyme. Soil sample was collected aseptically from Birnin Kebbi Fadama Farm, from a depth of 3cm with help of a sterile spatula, and placed in sterile plastic bag. The soil sample was immediately transported to the laboratory, and weighed for serial dilution. After serial dilution, 0.1ml of each dilutions (10-1, 10-3 and 10-5) were inoculated on starch casein agar plates, the plates were incubated at 37 0C for 48 hours. Growth of microorganisms were observed and the isolated bacteria were identified. The identified isolates (streptomyces) were inoculated in production media for 48 hours with constant shaking of 250 rpm. After production period, the activity of the enzyme was determined as well as the optimum carbon sources and incubation period. Based on physical, microbial and biochemical properties displayed by isolated bacteria, clearly shows that, the isolated bacteria were identified as Streptomyces, and were capable of producing pectinase enzyme with activity 0.213U/mol for the control and 0.347U/mol for the test. According to this finding the best carbon sources was sucrose and incubation period was after 36 hours.  The streptomyces obtained from Birnin kebbi fadama farmland were capable of producing pectinase enzyme which can be used in various industries.
